[2021-11-25 08:16:44] [Andrew Rambaut] This evening I think we may be back to square one with respect to vaccination and prior immunity. New pandemic anyone? [2021-11-25 08:22:26] [Robert Garry] I can model it - not possible to get a structure. It has probably 5 TM domains, which you expect of a membrane remodeler. The TMpred prediction is underlined and I bolded the "by-eye" most likely TMs. There are three internal domains that are in blue and italicized. There is a large external domain that has several cysteines -dicysteines - it's an uneven number so maybe a metal binding spot. The third external domain has a lot of aromatic amino acids - so does the putative fifth TM domain so that's probably the business part of the membrane remodeling. The T492I mutation is shown - it's at the end of the molecule - inside - 4 cysteines that domain could be regulatory. [2021-11-25 11:19:29] [Kristian Andersen] @Andrew Rambaut - I hope you're wrong, but I have been saying that for months - we're just one monster variant away, and maybe this is it... I bloody hope not - but just looking at the mutations, I would be _very_ surprised if an updated vaccine wouldn't be necessary. Not exactly square one, but it ain't great - plus, this is ignoring potential concerned about antigenic sin and/or ADE, but I don't want to think about either of those right now. [2021-11-25 12:13:22] [Kristian Andersen] Let's hope we're just looking at a sampling bias here - because if we're not, it doesn't matter what we do next. [2021-11-25 12:18:55] [Kristian Andersen] Let's hope not Bob. I think Nu could likely be a big problem, but surely the early data can't be correct. [2021-11-25 12:23:57] [Robert Garry] Nu - sorry - @Andrew Rambaut put Monty Python into my head and it's going to be hard to get away from that image of the knights... [2021-11-25 12:26:23] [Eddie Holmes] That graph and it's header is a bit irresponsible. Can't make those conclusions with focused sampling on a poorly immunized population. That said, I hope that Moderna and Pfizer have redesigned their vaccines already [2021-11-25 12:28:55] [Eddie Holmes] Both Andrew and I know Sean Nee well. He was not a fan of Monty Python. [2021-11-25 12:29:56] [Andrew Rambaut] It was estimated today that it would take 6-9 months to get an updated mRNA vaccine to deployable stocks. [2021-11-25 12:33:37]
