And of course optimal cleavage may not have peak fitness benefit to the virus - you can presumably cleave too well and alter some other trade off in the dynamics.

@Andrew Rambaut Agree with this - optimal cleavage might vary depending on multiple factors - cleave in the wrong tissue or wrong time not good. - An optimal cleavage site is RRXRR. Not all X's equal so RRRRR among the best.
