"In addition, nucleic fragments of SARS-CoV-2 were also detected using PCR and sequencing approaches. For
example, SARS-CoV-2 in wastewater in northern Italy using both nested RT-PCR targeting the ORF1ab region and
real-rime RT-PCR targeting the RdRp region, and the earliest positive samples were dated back to December 18,
2020 in Milan and Turin [La Rosa G et al., Sci Total Environ]. Similarly, the detection of SARS-CoV-2 in wastewatersamples was also reported using real-time RT-PCR targeting the N gene, with the archived sample collected inSpain on March 12, 2019 [Randazzo et al., Water research]. The SARS-CoV-2 RNA was also detected in a patientin France in December 2019 using two different PCR-based assays and his last international travel was to Algeria inAugust 2019 [Deslandes et al., IJAA]. However, none of the three studies reported the viral sequences from the positive samples. More recently, SARS-CoV-2 RNA was identified in an oropharyngeal swab specimen collected from a child with suspected measles in Italy in early December 2019 [Amendola et al., EID]. In addition, the amplified470-bp fragment of the spike gene from the child showed 100% identical to that of SARS-CoV-2. All these studies highlight the potential cryptic spread of SARS-CoV-2 in some European countries before the first reported case inrespective country, whose implications for the origins of SARS-CoV-2 clearly require further investigation."
