P.S. I did see the recent JCI Insight paper that shows impressive results for in vivo gene editing of the fetal globin promoter in the mouse model of sickle cell disease. Cites support from NHLBI, NHGRI, and NIGMS. Also support from BMGF. Cool stuff. See attached.

Hi all, Bill Gates is at the White House tomorrow and asked to see me at 9 AM. Among the topics we will likely discuss is the HIV-SCD joint NIH-BMGF effort to develop cures that will be applicable in low income settings. Are there any new developments since the Sunnylands meeting back in May? Bill will likely ask about the future of this partnership. Thanks, Francis

Hi Francis, On the DBDR side, we recently hosted Dr. Andre Lieber (the last author of the JCI manuscript) to present his work in this area as part of our CureSci initiative at NHLBI-agree very exciting. I continue to meet with Mike McCune and his team on SCD and am also participating in the SCD TPP activity that is progressing slowly. Thanks, Julie Julie A. niaid.nih.gov> Francis, We have continued to meet with Mike and his team. One of his team members is coming for a visit in October so we can have a face to face deep dive. Since Sunnylands the Temple group initiated the phase one of a anti-HIV CRISPR trial in Philadelphia, part of the NIAID Collaboratory. We have reviewed a set of grants looking new and improved delivery systems for gene therapies. We plan to fund 3-4 and the details will be announced in the new fiscal year. We are expanding in 2023 the process for sites in LMICs to apply for assistance to support cure trials including but not limited to gene-based technologies. The first iteration of this strategy has been completed, now on to version 2.0. Last, the plans for an bNAb treatment and cure trial in Africa with BMGF is moving forward. Waiting for BMGF to finish production of the bNAbs. This continues to be parallel and complementary activities with BMGF. Best regards, Carl
