It will be interesting to see what the Q677P mutation does to the O-glycans - it actually jacks up the prediction for each of the 3 sites quite a bit.
-
And 681R removes the O-glycan and makes it more basic?
-
How would you rank 677P, 681R and 681H?
-
Q677P - highly likelihood of 3/3 sites O-glycosylated in mucosal tissues. Q677H - only 2/3 sites with O-glycans -moderate probability that sites are used in mucosal tissues. 681R and H also knock down to only 2/3 sites with O-glycans - moderate probability that sites are used in mucosal tissues. Maybe "epistasis" going on with mutations like DouG that loosen-up the trimer and make it more "cleavable" - Maybe some of the FCS mutations tuning up the spot for optimum efficiency and timing of cleavage (like Goldilock's soup - not too hot not too cold).
-
Thanks!

681R is interesting in this regard - might lose some control over the cleavage, but overall make it more 'cleavable'?
