   Very important point - IF you were engineering in a FCS why use an out-of-frame insertion, add a freaking proline infront (unless you somehow were aware that that would help create the mystical O-linked glycans] and put in aminimal/suboptimal FCS (RXXR, instead of the optimal RXRR)? PooH and PooR are still trying to "get it right."

   I should say that 'suboptimal' in terms of cleavage may be optimal in terms of transmission (i.e., getting the right level of viral shedding in the right tissues at the right time given the behaviour of the human population) . But no way someone would be able predict that and until you tried it you would never know.
